Decipher cellular processes with real-time measurements
Ever been curious about internalization rates, mechanisms and how surrounding conditions may affect? In a recent paper, Encarnação and co-authors study the binding and internalization properties of a drug carrier, implementing a plethora of LigandTracer applications.
By following the drug carrier association to live cells at different temperatures, it was possible to understand how the binding and the internalization contributed to different phases of the curve. Once it had been established which part of the curve reflects internalization, the internalization rate could be quantified and it was found to be greatly influenced by the receptor saturation level and thus by the ligand concentration in solution. By implementing our proximity assay, receptor clustering could be followed in real-time to better understand the internalization mechanism. Both adherent and anchored suspension cells were used in the study.
